The series features both original and new voice actors. Sasha Knight voices Finn the Human, and John DiMaggio voices Jake the Dog. DiMaggio also voiced Jake the Dog in the original "Adventure Time" series, which aired on Cartoon Network from 2010 to 2018.

Other voice cast members for "Adventure Time: Side Quests" include Fred Tatasciore as Mannish Man, Jeff Bennett as Choose Goose, Hynden Walch as Princess Bubblegum, Aziah Pesaleli as Boyish Boy, Tom Kenny as Ice King, Olivia Olson as Marceline, and Niki Yang as BMO. The first episode of the series is titled "Bros and Arrows."

Cartoon Network Studios produced "Adventure Time: Side Quests." In an interview, John DiMaggio said, "It's got such a huge, huge fan base. There's such a large audience for this show. I don't know whose idea it was to bring the show back, but they're a genius." He added, "I really enjoy doing this part. I love to do it. I love the people that I work with. You know, because when you work on a show for a while, it becomes a family. Just a great bunch of people that are producing the show, writing the show, directing the show. It's exciting to come back and to do something again."

Knight described his connection to the role in an interview, saying, "I was a fan of the show growing up, and I watched it a lot when I was like pretty young because I have an older brother and he was a fan and he watched it. So it was always kind of on in our house. I'm just so happy that I get to bring Finn back to life." Regarding his performance, Knight stated, "I'm very similar to him in a lot of ways. And so it comes pretty naturally to me to just be Finn." He also commented on the show's appeal: "What's really special about this show is that it kind of can cater to everybody. If you've never seen the show and you're like a young fan and you want to start watching it, I feel like it's really great because it's very fun and goofy. And then if you've seen the show and you want more, then there's that too."
